Barcode construct for P. fluorescens knock-in

Barcode construct for P. fluorescens knock-in. Barcoded contains a barcode with a Rhamnose promoter upstream, for barcode transcription upon rhamnose inudction to trigger kill-switch mechanism. These two are flanked by strong terminators T1 and T7 to inhibit any unwanted transcription of the barcode. All of this if then flanked by two homology arms in either side for knock-in insertion attT7N trnasposon site of the gmlS gene found in the genome of prokaryotes. Barcode (BBa_K4663000) could be substituted with any other wanted barcode.
